Abstract: The present invention relates to fast disintegrating tablets of Domperidone formulated with chitosan-glycine conjugates as a superdisintegrant. Chitosan-glycine conjugates were prepared by physical mixing and microwave-assisted methods in different ratios and evaluated for flow, swelling, water uptake, effective pore radius, and related micromeritic properties. The conjugates were characterized using ATR-FTIR, X-ray diffraction, and scanning electron microscopy. Domperidone tablets were prepared by direct compression and evaluated for hardness, friability, weight variation, drug content, wetting time, water absorption, porosity, disintegration, and in vitro dissolution. Microwave-prepared conjugates demonstrated improved swelling, pore characteristics, and disintegration performance compared with pure chitosan and physical mixtures. The optimized formulation showed rapid disintegration and a dissolution profile comparable with the marketed product. The invention therefore provides a simple, effective, and patient-friendly fast disintegrating Domperidone tablet employing chitosan-glycine conjugates to improve tablet disintegration, drug release, and ease of administration, especially for patients with swallowing difficulties and enhanced treatment convenience.
