Abstract: The present invention relates to a fast disintegrating tablet formulation of esomeprazole magnesium (20 mg) prepared by the direct compression technique, employing superdisintegrants crospovidone or croscarmellose sodium at 10% to 20% w/w concentrations, along with microcrystalline cellulose, mannitol, sodium bicarbonate, colloidal silicon dioxide, talc, and magnesium stearate. Six formulations (F1 to F6) were developed and evaluated for pre- and post-compression parameters. The optimized formulation F6, containing croscarmellose sodium at 20% w/w, exhibited a disintegration time of 24 seconds, drug content of 99.9%, and in vitro dissolution of 95.1% at 30 minutes in pH 6.8 phosphate buffer. Accelerated stability studies at 40 degrees Celsius and 75% relative humidity for three months confirmed the physical and chemical stability of the product. The invention provides a cost-effective, patient-compliant solid dosage form for esomeprazole suitable for elderly, paediatric, and dysphagic patients.
