Abstract: The present invention relates to a formulation and method for preparing rutin-loaded oral microbeads comprising a sodium alginate polymeric matrix crosslinked with calcium chloride through an ionic gelation technique. The microsphere formulation system encapsulates rutin, a bioflavonoid antioxidant, within calcium alginate microbeads to achieve controlled and sustained oral drug release, improved bioavailability, and enhanced gastrointestinal stability. The microbeads are prepared by dispersing rutin in a sodium alginate solution and extruding the dispersion dropwise into a calcium chloride crosslinking bath under controlled stirring conditions. The resulting microbeads exhibit spherical morphology, high drug entrapment efficiency of up to 85.4 percent, pH-responsive swelling behaviour, and sustained drug release over 24 hours across varying physiological pH conditions. The invention provides an effective oral delivery system for bioflavonoid compounds with poor aqueous solubility and limited oral bioavailability.
