Abstract: The present invention discloses a biodegradable polymer-based hydrogel formulation for use as a controlled drug delivery system and a method for the preparation and characterization thereof. The hydrogel comprises an aqueous blend of chitosan, sodium alginate, and poly(ethylene glycol) diacrylate, ionically crosslinked using calcium chloride and covalently stabilized using genipin, a non-cytotoxic crosslinker. The therapeutic agent is loaded into the polymer blend prior to crosslinking to obtain uniform dispersion within the three-dimensional matrix. The hydrogel exhibits an equilibrium swelling ratio of three hundred and fifty to seven hundred and twenty per cent, drug entrapment efficiency exceeding eighty-five per cent, sustained release exceeding twenty-one days following near-zero-order kinetics with minimal burst, and controlled biodegradation over twenty-eight days. The formulation is biocompatible, mucoadhesive, scalable, and suitable for parenteral, topical, ocular, and oral controlled drug delivery applications.
