Abstract: The present invention provides a multifunctional rare earth ion-doped nanoparticle composition for fluorescence imaging in the second near-infrared window (1000 to 1700 nm) and targeted cancer therapy. The nanoparticle comprises a crystalline host matrix selected from NaYF4, KNdLiF, NaGdF4 or CeO2, doped with trivalent lanthanide ions such as Nd3+, Yb3+, Er3+ or Tm3+, with an inert or active shell layer for enhanced luminescence, a biocompatible polymeric coating for aqueous dispersibility, and surface-conjugated tumor-targeting ligands such as folic acid or antibodies. The nanoparticle exhibits tunable up-conversion and down-conversion emission, enables deep-tissue NIR-II imaging with tumor-to-background ratio exceeding ten, supports photodynamic therapy via reactive oxygen species generation, and provides stimuli-responsive drug release. The platform integrates diagnostic imaging, magnetic targeting and therapeutic action in a single nanoparticle architecture for advanced cancer theranostics.
