Abstract: The present invention relates to the field of peptide therapeutics and, more particularly, to novel therapeutic peptides capable of restoring intestinal epithelial barrier integrity through modulation of Zonulin-mediated tight junction dysfunction. The invention provides peptides comprising amino acid sequences selected from SEQ ID NOs: 1–15, pharmaceutical compositions comprising the peptides, methods of preparing the peptides using Fmoc-based solid-phase peptide synthesis, and methods of treating disorders associated with intestinal hyperpermeability. The disclosed peptides exhibit high-affinity binding to Zonulin, restore epithelial barrier function, reduce paracellular permeability, and promote tight junction integrity. The peptides are useful in the prevention and treatment of intestinal barrier disorders including inflammatory bowel disease, Crohn's disease, ulcerative colitis, and other diseases associated with epithelial barrier dysfunction.
