Abstract: A composition comprising an Azadirachta indica flower extract in combination with a standard therapeutic agent for polycystic ovary syndrome, and methods of preparing, evaluating, and using it, are disclosed. An extract of neem flowers, prepared by solvent extraction and standardized to a phytoconstituent, such as a limonoid or flavonoid, responsible for anti-polycystic-ovary-syndrome activity, is combined with a standard agent selected from an insulin sensitizer such as metformin, an anti- androgen, an ovulation inducer, and a hormonal agent, in proportions such that the combination exhibits an anti-polycystic-ovary-syndrome effect, amelioration of hyperandrogenism, insulin resistance, ovulatory dysfunction, and polycystic ovarian morphology, enhanced relative to the extract or the agent alone, or attained at a reduced dose of the agent with reduced adverse effects. The extract and agent act by complementary or synergistic mechanisms, the extract's antidiabetic, antioxidant, anti-inflammatory, and endocrine-modulating activities addressing aspects the agent does not. A method evaluates the potential by administering the combination, extract, and agent to models of the syndrome and comparing effects, establishing the enhanced or dose-sparing benefit for improved combination therapy.
