Abstract: The present disclosure relates to engineered nucleic acid sequences and recombinant vector constructs for coordinated overexpression of a miR-127/136 microRNA (miRNA) cluster. The engineered sequence comprises multiple miRNA stem-loop sequences arranged in a defined, non-naturally occurring configuration within a single contiguous nucleic acid sequence, optionally separated by spacer sequences. The construct is incorporated into an expression vector comprising regulatory elements including a promoter and a selectable marker. Methods are disclosed for introducing the construct into mammalian cells via viral or non-viral delivery systems and selecting stable cell lines expressing the miRNA cluster. The disclosure further provides engineered cell lines, screening platforms, and kits useful for functional studies of miRNA-mediated gene regulation and cancer research applications.
