Abstract: ABSTRACT ENGINEERED GENETIC CONSTRUCT COMPOSITIONS FOR IMMUNE MODULATION AND TRANSPLANT TOLERANCE The instant disclosure provides engineered immune-modulator construct compositions for localised, targeted, and inflammation-gated suppression of pathological immune activation. A library of sixty-one constructs (SEQ ID NOs: 1 to 61; FIGS. 1 to 61) is organised by architecture into six classes: exosome-displayed depots presenting PD-L1, interleukin-10, interleukin-4, TIGIT, transforming growth factor beta 3, or CD39/CD73 cargo; tissue-targeted constructs bearing chemokine-receptor or collagen-sensing homing cassettes (CCR7, CCR4, CXCR5, CX3CR1, CCR9, CCR2B, DDR2); inflammation-gated tolerogenic switches in which a serum-response- element promoter or a programmed death-1 cytoplasmic-domain substitution drives a tolerogenic output upon sensing inflammation; graft-localised immune-shielding constructs expressing HLA-G or PD-L1; dual-input Gal4-UAS combination constructs; and receptor-fusion constructs joining a homing receptor to a transforming- growth-factor-beta-receptor-2 module. Each construct is deployable across vesicular, nucleic acid, viral, and cellular presentation formats, and any application-context assignment is non-restrictive. (
