Abstract: ABSTRACT ENGINEERED EXOSOMAL DNA CASSETTES AND SCAFFOLD FOR SPINAL CORD AND AXON REGENERATION The instant disclosure provides a concoction of 51 engineered DNA cassettes (SEQ ID NOs: 1 to 51) for spinal cord and axon regeneration, classified into cytoprotective and oxidative-stress, immune-modulation, pathology-driver silencing, growth-transcription-factor, partial-reprogramming, neurotrophic-ligand, growth-oriented-receptor, guidance, remyelination, pathway-antagonist and molecular-movement modules, the cargo loaded into or displayed upon engineered extracellular vesicles by a multi-pathway sorting-motif architecture. Reversible dCas9-KRAB repressors release the growth brakes PTEN and suppressor-of-cytokine-signalling-3 and de-repress EphA4- mediated guidance; KLF-6, SOX11 and osteopontin drive the regenerative state; Netrin-1 and GAP-43 guide the regrowing axon; a kinesin motor domain (SEQ ID NO: 51) supports anterograde transport; an OLIG2 remyelination module is deferred to a delayed-release fraction following axonal traversal; and each neurotrophic ligand is co-presented with its cognate receptor. The instant disclosure further provides an electrically semi-conductive hydrogel micro-thread scaffold article incorporating the vesicle population along a spatial gradient and a temporally gated delayed-release sequence, and methods of manufacture thereof.
