Abstract: ABSTRACT ENGINEERED EXOSOMAL DNA CASSETTES AND SCAFFOLD FOR SCHWANN-CELL AND PERIPHERAL NERVE REPAIR The instant disclosure provides a concoction of 45 engineered synthetic DNA cassettes (SEQ ID NOs: 1 to 45) for Schwann-cell and peripheral nerve repair, classified into cytoprotective and oxidative-stress, immune-modulation, pathology-driver silencing, partial-reprogramming, neurotrophic-ligand, growth-oriented-receptor, guidance, pathway-antagonist and molecular-movement modules, the cargo of which is loaded into or displayed upon engineered extracellular vesicles by a multi-pathway sorting-motif architecture. A reversible, non-integrating dCas9-KRAB repressor reduces peripheral myelin protein-22 gene dosage in Schwann cells toward a physiological level without permanent ablation; a partial-reprogramming module is delivered through a prolyl-endopeptidase and thimet-oligopeptidase cleavable linker; each neurotrophic ligand is co-presented with its cognate receptor; and a kinesin motor domain (SEQ ID NO: 45) restores axonal transport. The instant disclosure further provides an electrically semi-conductive hydrogel micro-thread guidance conduit incorporating the engineered extracellular-vesicle population along a bilateral exponential spatial gradient and a temporally gated delayed-release sequence, and methods of manufacture thereof.
