Abstract: ABSTRACT ENGINEERED DNA CASSETTE WITH AN INSULATOR ENCODING A MODIFIED GLOBIN WITH INTEGRATED SAFETY-SWITCH The instant disclosure provides an engineered DNA cassette (100) comprising a promoter (112), a Kozak sequence (114), a modified globin coding sequence (116) carrying an anti-polymerization T88Q substitution (118), a T2A self-cleaving peptide element (120) and a safety-switch module (122). The modified globin and the safety- switch module (122) are encoded as a single open reading frame resolved co-translationally by the element (120), whereby the safety-switch module (122) is co- expressed in fixed stoichiometric linkage with the modified globin and renders the carrying cell selectively eliminable. A 77 bp insulator sequence (128) segregates the open reading frame from a Bleomycin selection marker (138) to suppress promoter interference. The safety-switch module (122) is interchangeably a surface depletion tag (122a; SEQ ID NO: 1), a thymidine kinase element (122b; SEQ ID NO: 2) or a yeast cytosine deaminase-UPRT fusion (122c; SEQ ID NO: 3).
