Abstract: The invention provides protein based, DNA-based, and protein nanoparticle vaccines useful in the prevention of Dengue and/or Zika virus infection. The vaccines comprise engineered dimeric envelope glycoproteins derived from consensus envelope sequences of dengue virus serotypes 1-4 and Zika virus. The envelope glycoproteins are designed to form stable dimers that present native-like quaternary epitopes, including envelope dimer epitopes associated with broadly neutralizing antibody responses. The disclosed vaccine compositions elicit robust and cross-reactive humoral immune responses while minimizing the induction of non-neutralizing or infection-enhancing antibodies. The invention further provides methods of inducing protective immunity against dengue and/or Zika virus in a subject by administering the disclosed vaccine compositions. Fig. 4
