Abstract: The present invention relates to an electrospun nano-fibrous transderalensional transdermal patch comprising Tofacitinib citrate incorporated. dissolved within a polymer matrix for sustained anti-arthritic therapy. The patch is fabricated using Eudragit polymer and a solvent system comprising ethanol and N,N- dimethylformamide. The drug-loaded polymer solution is subjected to electrospinning under optimized conditions to produce uniform, bead-free nano-fibers having high surface area and enhanced transdermal permeation properties. The fabricated patch exhibits controlled and sustained release of Tofacitin over a period of about 24 hours with high drug content uniformity and stability. Characterization studies confirm compatibility between the drug and excipients and demonstrate nano-scale fiber morphology. The transdermal therapeutic system provides prolonged drug release, improved therapeutic efficacy, reduced systemic adverse effects, and enhanced patient compliance for management of rheumatoid arthritis and related inflammatory disorders.
