Abstract: The present invention relates to a dual-polymer controlled-release matrix tablet comprising pirfenidone and vericiguat for sustained drug delivery. The formulation includes a hydrophilic matrix of hydroxypropyl methylcellulose (HPMC K100M) and high-molecular-weight polyethylene oxide (PEO) in a weight ratio of about 3:1 to 1:1, preferably 2:1, at a total polymer content of about 9–11% w/w. Poloxamer 188 is incorporated to enhance the dissolution of vericiguat, while caprylic acid, present at about 22% by weight, promotes faster dissolution and provides synergistic therapeutic effects. The interpenetrating dual-polymer matrix forms a gel-diffusion barrier that compensates for the differing solubility profiles of the active ingredients and enables substantially synchronized, near zero-order drug release for at least 12 hours. The tablet is prepared by aqueous wet granulation followed by compression, providing a stable, scalable fixed-dose combination with reduced dosing frequency, improved patient compliance, and suitability for conventional pharmaceutical manufacturing.
