Abstract: The present invention relates to dual drug-loaded lipid-polymer hybrid nanoparticles for the management of diabetic nephropathy. The nanoparticles are prepared by a nanoprecipitation-emulsification method using PLGA as the polymeric core, phospholipids and cholesterol as the lipid shell components, and a suitable surfactant for stabilization. The formulation co-encapsulates two therapeutically complementary drugs targeting oxidative stress and inflammatory pathways. The optimized nanoparticles exhibit a particle size of 168.5 nanometers, polydispersity index of 0.21, zeta potential of negative 24.6 millivolts, and entrapment efficiency of 88.0 and 85.0 percent for Drug 1 and Drug 2 respectively. The formulation demonstrates controlled and sustained drug release over 24 hours and provides superior nephroprotective activity with improved cellular viability, reduced oxidative stress, and attenuated inflammation in diabetic nephropathy models compared to free drugs.
