Abstract: ABSTRACT The present invention relates to a dual-action antibacterial–antifungal mucoadhesive hydrogel for localized management of oral infections. The developed hydrogel comprises at least one antibacterial agent, at least one antifungal agent and a biocompatible mucoadhesive polymeric matrix. In a preferred embodiment, the formulation comprises chlorhexidine as an antibacterial component and clotrimazole or nystatin as an antifungal component incorporated into a carbomer-chitosan hydrogel matrix. The optimized formulation demonstrated suitable pH, viscosity, spreadability and mucoadhesive strength and remained associated with oral mucosal tissue for an extended period under simulated salivary conditions. In-vitro studies demonstrated controlled and sustained release of both antimicrobial components. The formulation exhibited significant antibacterial activity against representative oral bacterial pathogens and antifungal activity against Candida species. The optimized hydrogel further demonstrated substantial inhibition of bacterial, fungal and mixed microbial biofilms while maintaining acceptable cytocompatibility. The invention provides a single localized dosage form capable of simultaneously addressing bacterial and fungal components of oral infection while prolonging antimicrobial residence at the oral mucosal surface.
