Abstract: A microbiome-responsive system (100) for the targeted topical delivery of therapeutic agents in the treatment of inflammatory skin disorders associated with microbial dysbiosis. The system (100) comprises a core matrix (101) having a particle size of 50-500 nm and encapsulating a therapeutic payload (102) selected from anti-inflammatory phytochemicals, anti-staphylococcal agents, or probiotics. A responsive coating (103) envelops the core and incorporates a sensor element (104) designed to specifically recognize a pathogenic microbial biomarker (104a) such as a quorum-sensing peptide or protease. Upon binding of the biomarker (104a), the sensor element (104) triggers structural destabilization or cleavage of a responsive linker, inducing localized and on-demand release of the payload (102). This adaptive release mechanism enables precise site-specific therapy, minimizes off-target effects, and supports restoration of microbiome balance in conditions such as atopic dermatitis and other dysbiosis-related skin inflammations.
