Abstract: The present invention discloses a dissolving microneedle patch composition comprising tapinarof and dextran-coated cerium oxide nanozyme dispersed within a polymeric matrix of polyvinylpyrrolidone and polyvinyl alcohol for synergistic transdermal amelioration of psoriasis. The microneedle patch bypasses the thickened psoriatic stratum corneum and delivers both agents directly into the epidermis and dermis. Tapinarof activates the aryl hydrocarbon receptor to downregulate pro- inflammatory cytokines including interleukin-17 and upregulates endogenous antioxidant defences through the Nrf2 pathway. The cerium oxide nanozyme provides direct, catalytic, and self-regenerating scavenging of reactive oxygen species through superoxide dismutase-like and catalase-like activities via Ce3+/Ce4+ redox cycling. The combination exhibits synergistic ROS scavenging and anti-inflammatory effects. In vivo studies in an imiquimod-induced psoriasis mouse model demonstrated significantly superior PASI score reduction for the combination microneedle patch compared to individual agents. The invention also discloses the process for fabricating the microneedle patch.
