Abstract: Dispersible Tablet Formulation of Itraconazole for Enhanced Solubility and Bioavailability This invention describes a dispersible tablet formulation designed to enhance the solubility, dissolution, and bioavailability of Itraconazole, a poorly water-soluble antifungal drug. Utilizing solid dispersion techniques with hydrophilic carriers, such as PEG 4000 and PEG 6000, the drug is stabilized in an amorphous state to improve its aqueous solubility and dissolution rate. Super-disintegrants, including Crosspovidone, Crosscarmellose Sodium, and Sodium Starch Glycolate, ensure rapid tablet disintegration in under two minutes upon contact with water, forming a homogenous suspension for ease of administration. The optimized formulation (F3) achieves 90.71% cumulative drug release within 45 minutes, ensuring faster therapeutic onset and consistent absorption irrespective of food intake. Scalable through direct compression methods, this innovation provides a patient-centric, cost-effective solution for antifungal therapy, addressing key challenges such as swallowing difficulties and variable bioavailability, while maintaining compliance with pharmaceutical manufacturing standards.
