Abstract: ABSTRACT TITLE: DIMETHYL PYRROLE-BASED TRIAZOLOTHIADIAZOLE DERIVATIVES AS ANTI-TUBERCULAR AGENTS AND A PROCESS FOR THE PREPARATION THEREOF The present invention discloses novel dimethyl pyrrole-based [1,2,4]triazolo[3,4-b][1,3,4]thiadiazole derivatives (SP-5A to SP-5P) and a process for their preparation and use as anti-tubercular agents. The compounds are prepared by condensing 4-amino-5-[4-(2,5-dimethyl-1H-pyrrol-1-yl)phenyl]-4H-1,2,4-triazole-3-thiol with substituted benzoic acids in phosphorus oxychloride, introducing diverse aromatic substituents at the 6-position. The synthesised compounds, characterised by FT-IR and 1H NMR spectroscopy, were screened against Mycobacterium tuberculosis H37Rv by the Microplate Alamar Blue Assay. Compounds SP-5K and SP-5M exhibited superior activity with a minimum inhibitory concentration of 12.5 microgram per millilitre, while several analogues showed moderate activity at 25 microgram per millilitre. The invention demonstrates that appropriate aromatic substitution on the pyrrole-triazolothiadiazole framework enhances antimycobacterial potency, providing promising lead molecules for the development of new therapies against drug-resistant tuberculosis.
