Abstract: ABSTRACT [0033] The present study aimed to develop and optimize a Gentamicin loaded nanoemulgel for enhanced topical drug delivery in the management of bacterial skin infections. The formulation was designed to improve skin permeation, provide sustained drug release, enhance localized antibacterial activity, and reduce systemic exposure associated with conventional therapy.Preformulation studies, including solubility analysis, UV spectrophotometric method, calibration curve, and FTIR compatibility studies, were performed to evaluate the suitability of the drug and excipients. Nanoemulsions were prepared using optimized oil, surfactant, co- surfactant, and distilled water based on pseudo-ternary phase diagram studies. The optimized nanoemulsion was incorporated into a Carbopol 940 gel base to prepare the nanoemulgel. [0034] The developed formulations were evaluated for particle size, polydispersity index, zeta potential, SEM, drug content, pH, viscosity, spreadability, extrudability, in vitro drug release, release kinetics, antibacterial activity, and stability studies. The optimized nanoemulgel exhibited nanosized droplets with uniform size distribution, acceptable polydispersity index, satisfactory zeta potential, high drug content, suitable pH, optimum viscosity, good spreadability, and excellent extrudability. The formulation demonstrated sustained drug release, followed favorable release kinetics, showed effective antibacterial activity, and remained physically stable throughout the stability study. The optimized gentamicin sulfate-loaded nanoemulgel demonstrated desirable physicochemical characteristics, sustained drug release, effective antibacterial activity, and good stability. These findings suggest that the developed formulation has potential as an efficient topical drug delivery system for the treatment of bacterial skin infections with improved therapeutic performance and reduced systemic adverse effects.
