Abstract: The present invention discloses a statistically optimised nutraceutical nanoparticulate delivery system in which L-arginine is encapsulated within a chitosan- tripolyphosphate polyelectrolyte matrix prepared by ionic gelation. The formulation is developed through a face-centered central composite design that simultaneously varies polymer concentration, cross-linker concentration and arginine loading at three coded levels, and yields quadratic response surface models for particle size, zeta potential and encapsulation efficiency with adjusted coefficients of determination above 0.97. The optimised composition provides nanoparticles of about 240 nanometers, zeta potential of +54 millivolts, encapsulation efficiency of 81 percent and polydispersity index below 0.30. In vitro dissolution demonstrates sustained release of 86 percent over 12 hours, and in vivo studies in rats show a 2.7-fold enhancement in oral bioavailability compared with free L-arginine, together with excellent long-term and accelerated storage stability suitable for commercial nutraceutical use.
