Abstract: The present invention relates to a phytosome-loaded berberine transdermal patch for management of diabetes mellitus. The invention comprises berberine or a pharmaceutically acceptable salt thereof associated with at least one phospholipid to form a phytosomal delivery system, wherein the phytosomal berberine is incorporated into a polymeric transdermal matrix comprising one or more film-forming polymers, plasticizers and optionally penetration enhancers. The phytosomal system modifies the physicochemical characteristics of berberine and facilitates interaction with biological membranes, while the polymeric matrix provides controlled and sustained drug release. The transdermal patch is designed to improve skin permeation and systemic delivery of berberine while potentially reducing limitations associated with conventional oral administration. The formulation may be characterized for particle size, polydispersity, zeta potential, physicochemical compatibility, drug content, mechanical properties, in-vitro release, ex-vivo skin permeation, skin retention, stability and pharmacodynamic antidiabetic activity. The invention provides a potentially scalable transdermal delivery platform for improving the pharmaceutical performance of berberine in the management of diabetes mellitus.
