Abstract: ABSTRACT TITLE: DEVELOPMENT AND EVALUATION OF OMEPRAZOLE-LOADED FLOATING MUCOADHESIVE MICROSPHERES FOR PROLONGED ANTI-ULCER TREATMENT The present invention relates to an omeprazole-loaded floating mucoadhesive microsphere formulation for prolonged anti-ulcer treatment, prepared by a hybrid ionotropic gelation and emulsion solvent evaporation process using sodium alginate, chitosan, hydroxypropyl methylcellulose K4M and ethyl cellulose cross-linked with calcium chloride. The optimised microspheres, obtained at a polymer to drug ratio of two-to-one and a calcium chloride concentration of ten percent, exhibit a drug entrapment efficiency of about eighty-nine point five percent, a floating lag time of about forty-nine seconds, a total floating time exceeding twelve hours, in vitro mucoadhesion of about eighty-four point six percent on gastric mucosa, and sustained release over twelve hours following Korsmeyer-Peppas anomalous transport. The multi-particulate system synergistically combines buoyancy and mucoadhesion, provides localised sustained omeprazole delivery, improves therapeutic efficacy in peptic ulcer disease, permits once-daily dosing, and demonstrates excellent stability under accelerated storage conditions.
