Abstract: Endometriosis is a chronic estrogen-dependent inflammatory disorder causing pelvic pain and infertility. Current hormonal therapies suffer from significant adverse effects and high recurrence rates. The present invention discloses a sterile injectable formulation comprising standardized hydroalcoholic extracts of Withania somnifera (withanolides), Asparagus racemosus (shatavarins), Camellia sinensis (epigallocatechin gallate), and Zingiber officinale (gingerols) dissolved in a co-solvent system of ethanol, propylene glycol, and polysorbate 80 in water for injection. The formulation bypasses first-pass metabolism, achieving rapid systemic bioavailability and multi-target pharmacological action including anti-inflammatory, antioxidant, anti-angiogenic, and hormonal modulation activities. Preclinical evaluation in a rat model demonstrated 66.7 percent reduction in endometriotic lesion volume, 57.9 percent reduction in tumor necrosis factor-alpha, 64.0 percent reduction in interleukin-6, and 57.6 percent reduction in vascular endothelial growth factor at the high dose, with restoration of oxidative stress markers toward normal values, establishing the formulation as a promising non-hormonal therapeutic alternative for endometriosis management.
