Abstract: ABSTRACT The present invention discloses a novel anti-inflammatory topical herbal gel formulated using ethanolic leaf extract of Couroupita guianensis (Cannonball Tree; Lecythidaceae). Conventional synthetic anti-inflammatory preparations cause skin irritation, systemic toxicity, and dependency upon prolonged use. The invented gel utilizes the plant's phytochemical profile — alkaloids, flavonoids (quercetin, rutin, kaempferol), saponins, tannins, and terpenoids (ursolic acid, beta-sitosterol) — to deliver dual anti- inflammatory and antimicrobial activity topically. Carbopol 940 serves as the gelling agent, with propylene glycol, triethanolamine, parabens, and purified water as excipients. The optimized batch demonstrated a skin-compatible pH of 6.4, viscosity of 495,000 cPs, and spreadability of 3.0 g·cm/sec. In vitro protein denaturation inhibition confirmed anti-inflammatory activity at 300 µg/mL. Antibacterial efficacy against Staphylococcus aureus and controlled phytoconstituent release peaking at 45 minutes via Franz diffusion were established. Stability studies confirmed formulation robustness under ambient and refrigerated storage.
