Abstract: DEVELOPMENT AND CHARACTERIZATION OF PROTRANSFERSOMAL GEL FOR TRANSDERMAL DELIVERY OF ANTIRESORPTIVE DRUG ABSTRACT The present invention relates to the development and characterization of a protransfersomal gel for the transdermal delivery of antiresorptive drugs, particularly for the management of osteoporosis. Conventional oral administration of antiresorptive agents is often associated with poor bioavailability, gastrointestinal adverse effects, and reduced patient compliance. To overcome these limitations, a protransfersomal drug delivery system was designed using phospholipids and an edge activator to enhance drug permeation through the skin. The protransfersomal vesicles were prepared and incorporated into a suitable gel base to obtain a stable transdermal formulation. The developed system was evaluated for vesicle size, morphology, entrapment efficiency, drug content, in vitro drug release, skin permeation, and stability. The optimized formulation demonstrated high drug encapsulation, improved deformability, and enhanced transdermal permeation compared with conventional gel formulations. Stability studies confirmed the physicochemical integrity of the formulation during storage. The findings indicate that the protransfersomal gel provides a promising non-invasive alternative for the sustained and effective delivery of antiresorptive drugs, potentially improving therapeutic efficacy, reducing systemic side effects, and enhancing patient adherence in osteoporosis treatment.
