Abstract: The current invention is concerned with the development of novel coumarin-clubbed thiazolidinediones that exhibit potential antibacterial and antifungal properties. The invention concerns the development of hybrid heterocycles of general Formula I where coumarin and thiazolidinedione pharmacophores are bonded to each other through an acetate bridge and then further modified by benzylidene groups. The developed compounds have been synthesized by multistep procedure that includes the formation of ester between 7-hydroxy-4-methyl coumarin and 2-bromoacetyl bromide, followed by the nucleophilic addition with thiazolidine-2,4-dione and the Knoevenagel condensation reaction with various benzaldehyde compounds using piperidine as catalyst. The developed compounds were characterized by ¹H-NMR and ¹³C-NMR. The antibacterial and antifungal activities of developed compounds were also studied against selected strains of Gram-positive bacteria, Gram-negative bacteria and fungi via in-vitro. Some of the synthesized compounds were found to possess promising antimicrobial activities similar to standard drugs.
