Abstract: The present invention discloses denatonium saccharide and denatonium benzoate compounds and an isocratic reverse-phase ion-pair high-performance liquid chromatography method for their simultaneous identification and quantitative assay. The method employs an octadecylsilane C-18 column of dimensions 4.6 mm by 250 mm packed with 5-micrometre particles, a binary mobile phase comprising an aqueous phosphate buffer containing 1-octane sulphonic acid sodium salt as ion-pair reagent and acetonitrile in a volume ratio of 50:50 with pH adjusted to 3.5 to 4.2, a flow rate of 1.0 mL/min, an injection volume of 10 microlitres, ultraviolet detection at 211 nm, and a total run time of ten minutes. The method achieves baseline resolution of the denatonium cation from the benzoate and saccharide counter-ions, is suitable for routine quality-control testing of bulk drug and finished consumer products, and is robust, economical and validatable.
