Abstract: Abstract The present invention relates to a curcumin-loaded niosomal aerosol formulation and a method for its preparation for targeted pulmonary drug delivery. The formulation comprises curcumin encapsulated within niosomal vesicles prepared using non-ionic surfactants, cholesterol, and pharmaceutically acceptable excipients to enhance the solubility, stability, and pulmonary delivery of curcumin. The preparation method involves dissolving the lipid components and curcumin in an organic solvent, forming a thin lipid film by solvent evaporation, hydrating the film with an aqueous buffer to produce niosomal vesicles, reducing vesicle size, stabilizing the formulation by lyophilization, and processing it into an inhalable aerosol suitable for administration through pulmonary delivery devices. The invention provides a reproducible and scalable manufacturing process for producing a stable aerosol formulation intended for localized delivery of curcumin to the lungs. The formulation is applicable for respiratory disorders requiring targeted pulmonary administration of anti-inflammatory, antioxidant, and antiviral therapeutic agents.
