Abstract: TITLE: CRISPR NANO-DETECTOR FOR ULTRA-SENSITIVE CTDNA CANCER BIOMARKER DETECTION USING NANOPARTICLE SIGNAL AMPLIFICATION APPLICANT: 1. MEENAKSHI ACADEMY OF HIGHER EDUCATION AND RESEARCH 2. MEENAKSHI AMMAL DENTAL COLLEGE AND HOSPITAL ABSTRACT The present invention discloses a circulating tumor DNA (ctDNA) CRISPR nano-detector system for ultra-sensitive cancer biomarker detection. The system comprises a ctDNA extraction module configured to isolate tumor-derived nucleic acids from blood or plasma samples, a CRISPR recognition unit comprising Cas12a and/or Cas13a enzymes complexed with mutation-specific guide RNAs, reporter probes configured to generate detectable signals upon CRISPR activation, a nanoparticle amplification unit comprising gold nanoparticles, quantum dots, and/or magnetic nanoparticles for signal enhancement, and a portable detection device configured for real-time signal analysis and diagnostic interpretation. The system performs ctDNA isolation, mutation-specific recognition, CRISPR-mediated reporter cleavage, nanoparticle-assisted signal amplification, and real-time analysis to enable rapid detection of cancer-associated biomarkers. The system provides high analytical sensitivity, multiplex biomarker detection, reduced background interference, portability, and suitability for point-of-care oncology diagnostics.
