Abstract: The present invention relates to a computer-implemented method and system for generating a Composite Pharmacogenomic Risk Score (CPRS) for pediatric epilepsy patients on sodium valproate therapy. The method integrates pharmacogenomic genotype data from UGT1A6, UGT2B7, UGT1A3, CYP3A4, and CYP2C9 single nucleotide polymorphisms with demographic, dosage, and longitudinal biochemical parameters including liver function tests, platelet count, and serum valproate concentration. Four independently trained machine learning models compute component scores for seizure recurrence (S1), hepatotoxicity (S2), thrombocytopenia (S3), and drug concentration adequacy (S4). These scores are normalized and aggregated as CPRS = (S1_norm + S2_norm + S3_norm + S4_norm)/4, producing a single metric ranging from 0 to 1. The system stratifies patients into low, moderate, or high risk categories and issues clinical recommendation flags, enabling individualized dose adjustment and therapeutic monitoring in pediatric valproate management.
