Abstract: COMPOSITE SPRAY-DRIED AMORPHOUS SOLID DISPERSION OF TICAGRELOR WITH ENHANCED DISSOLUTION AND MANUFACTURING PROCESS ABILITY The present invention relates to a spray-dried composite amorphous solid dispersion comprising ticagrelor, a poly (acrylic acid)-based polymeric carrier, preferably Apinovex®, and a porous inorganic excipient, preferably magnesium aluminometasilicate (Neusilin®), and to a process for its preparation. The dispersion is prepared by spray drying a homogeneous feed suspension in an ethanol–acetone solvent system, whereby ticagrelor is transformed from a crystalline state into a molecularly dispersed amorphous form. The polymeric carrier stabilizes the amorphous drug through molecular interactions, while the porous inorganic excipient adsorbs the drug–polymer matrix, reducing particle stickiness and agglomeration during spray drying. The resulting composite dispersion exhibits enhanced process yield, improved powder flowability, and superior dissolution performance compared with conventional crystalline ticagrelor and polymer-only amorphous dispersions. Solid-state characterization confirms successful amorphization, drug–polymer interactions, and formation of homogeneous composite particles. The invention provides a scalable and industrially applicable formulation platform suitable for oral solid dosage forms with enhanced manufacturability and bioavailability.
