Abstract: TITLE: CLOSED-SYSTEM DUAL-CHAMBER DEVICE FOR INSTANT REAGENT MIXING AND REAL-TIME BONE DECALCIFICATION ENDPOINT DETECTION APPLICANT: 1. MEENAKSHI ACADEMY OF HIGHER EDUCATION AND RESEARCH 2. MEENAKSHI AMMAL DENTAL COLLEGE AND HOSPITAL ABSTRACT The present invention discloses a closed-system pressure-activated dual-chamber device for detecting the endpoint of bone decalcification during histopathological processing. The device comprises a first chamber storing a 4% aqueous ammonium oxalate solution and a second chamber storing a 2% aqueous ammonium hydroxide solution, wherein the chambers are separated by a frangible barrier. A pressure-activation mechanism comprising a plunger is configured to rupture the frangible barrier and enable mixing of the reagents to form an active detection solution. The device further comprises a syringe-like housing body and an inlet configured to aspirate a sample of decalcifying solution into contact with the active detection solution. Residual calcium ions present in the sample react with the active detection solution to form a visible calcium oxalate precipitate, indicating incomplete decalcification, while absence of precipitate indicates completion of decalcification. The device enables reagent storage, on-demand activation, sample testing, and real-time endpoint determination within a sealed contamination-free platform suitable for histopathological applications.
