Abstract: A system (102) and a method (500) for closed-loop adaptive medication scheduling is disclosed. The system (102) includes wearable biosensors to continuously monitor physiological and motor signals of patient and transmit sensor data representative of motor state of patient. The system includes processors that are configured to receive sensor data and generate a patient-specific model predicting a pharmacological response of the patient to medication. The processors receive an estimated motor state derived from digital twin unit and determine an adaptive dosing schedule. The processors receive adaptive dosing schedule and generate actuation commands for one or more drug delivery devices, and evaluate the actuation commands against one or more safety constraints and modify or suppress the actuation commands upon detection of an adverse risk condition. The system operates as closed loop in which post-dose physiological feedback from wearable biosensors is used to continuously update reinforcement learning controller and digital twin unit.
