Abstract: The present invention relates to the field of medicinal chemistry, specifically to the development of novel compounds for inhibiting Plasmodium falciparum lactate dehydrogenase (PfLDH), a key enzyme in the malaria parasite's glycolytic pathway. The invention discloses compounds comprising chromone and thiochromone derivatives, which include a chromone or thiochromone scaffold modified with electron-donating groups (EDG) such as amine, hydroxyl, methyl, ether, and ethyl, and electron-withdrawing groups (EWG) such as nitro, chlorine, carboxyl, and aldehyde. These groups are strategically placed at specific positions on the chromone/thiochromone ring and a phenyl ring attached to a sulphonylpiperazine moiety, enhancing the compound's ability to bind and inhibit the PfLDH enzyme. The invention further encompasses methods for synthesizing these derivatives, utilizing molecular docking studies to predict binding affinity, molecular dynamics simulations to assess stability and interaction, and in silico ADMET predictions to evaluate pharmacokinetic properties.
