Abstract: The present invention discloses a nano-formulated therapeutic composition comprising whole garlic (Allium sativum) pod extract encapsulated within chitosan nanoparticles for use in the treatment or attenuation of high glucose induced kidney cell fibrosis. The nanoparticles are prepared by ionic gelation using sodium tripolyphosphate as a crosslinking agent and exhibit a hydrodynamic diameter of approximately 70–90 nm, a polydispersity index of approximately 0.171, and an encapsulation efficiency of at least 97%. The formulation provides a sustained and controlled release profile under physiological conditions without an initial burst release. Biological evaluation using NRK-49F renal fibroblast cells demonstrates biocompatibility at concentrations up to 60 µg/ml, significant inhibition of high glucose-induced cell proliferation (1.8–2.0 fold reduction) and migration (2.5–3.3 fold reduction), and attenuation of oxidative stress as evidenced by reduction in malondialdehyde (MDA) levels (1.7–2.0 fold reduction) under high glucose conditions. The present invention thus provides a simple, reproducible, and scalable nano-therapeutic approach for managing cellular mechanisms associated with renal fibrosis.
