Abstract: ABSTRACT CHIMERIC ANTIGEN RECEPTOR IMMUNE CELLS WITH PD-1 SIRNA, SUICIDE GENE AND INDUCIBLE THERAPEUTIC MODULES The instant disclosure provides engineered immune cells comprising a chimeric antigen receptor co- encoded with a constitutively expressed synthetic PD-1 silencing module and a selectable suicide gene safety system within a single lentiviral expression construct, wherein the chimeric antigen receptor incorporates an antigen-binding domain directed against CD19, glypican-3, Tn-MUC1, or Siglec family proteins for application across haematological and solid tumour indications. In additional embodiments, the engineered immune cell platform is generated through sequential transduction with three functionally distinct lentiviral constructs, the second encoding a constitutively expressed human sodium iodide symporter enabling PET imaging of tumour localisation and a reverse tetracycline-controlled transactivator, and the third encoding at least one therapeutic payload under doxycycline-responsive tetracycline response element control. In further embodiments, the inducible therapeutic payload comprises a C1C2 domain-fused molecule configured for extracellular vesicle-mediated tumour-local delivery. Lentiviral expression constructs and manufacturing methods are also provided. [
