Abstract: The disclosure relates in some aspects to mammalian retinal pigment epithelial cells comprising an engineered activatable polynucleotide encoding a master regulatory transcription factor, compositions comprising the cells, methods of producing the cells, and methods of treatment using the cells. The disclosure also relates in some aspects to retinal grafts comprising induced pluripotent stem cell (iPSC)-derived retinal pigment epithelial cells, a proportion of which comprise an engineered polynucleotide comprising an inducible promoter operably linked to a neurogenin-2 coding sequence. Methods of using the retinal pigment epithelial cells and grafts for treating eye diseases are also provided herein.
