Abstract: The ripe pod pulp of Cassia fistula linn. (Fabaceace) has long been used as a traditional laxative drug due to Quercetin, as the major flavanoid constituent in the pod pulp. It is widely used for treating hemorrhoids. In this study, methanolic extract from ripe pod pulp of Cassia fistula was studied for presence of phytochemicals and solvent system was developed for thin layer chromatography and further high performance thin layer chromatography was performed. Herbal suppositories were developed using extract of Cassia fistula linn. (Fabaceace) and evaluated for various parameters such as liquefaction, disintegration, melting point, weight variations, hardness test, in vitro dissolution and pharmacokinetic studies. Anti-oxidant potential of cassia fistula suppositories was performed. It was found that cassia fistula suppositories contain various plant secondary metabolites and HPTLC study confirmed the presence of Quercetin in large amount. The optimized suppositories based on in vitro dissolution pharmacokinetic study shows better anti-oxidant potential compared to pure extract of cassia fistula in DPPH assay so it may be concluded these herbal suppositories can be used for treatment of hemorrhoids.
