The application for the patent was published on August 28, 2026, under issue no. 35/2026.
Abstract: The present invention discloses an integrated microfluidic cartridge (100) designed for extraction-free direct Reverse Transcription Polymerase Chain Reaction (RT-PCR). The cartridge enables a rapid sample-to-answer workflow by integrating a Sample Inlet (110), a Lysis Chamber (130) utilizing thermal and chemical lysis, Reagent Storage Chambers (140) containing pre-loaded lyophilized reagents (142), and distinct Thermal Zones (170) for Reverse Transcription (150) and PCR Amplification (160) within a single disposable unit. The invention eliminates the need for conventional nucleic acid extraction and cold-chain reagent storage. The cartridge features an Optical Window (180) for real-time detection and utilizes a sealed architecture to ensure biosafety. The present invention includes passive fluidic valves and phase-change materials for robust thermal control, making the device suitable for Point- of-Care (POC) molecular diagnostics in resource-limited settings.
