Abstract: ABSTRACT CAPRINE AMNIOTIC MEMBRANE-BASED BIO-SCAFFOLD, BIOACTIVE HYDROGEL FORMULATION AND PROCESS OF PREPARATION THEREOF The present disclosure relates to a biological scaffold and a bioactive hydrogel formulation. The biological scaffold comprises a caprine amniotic membrane obtained from a full-term placenta post-parturition, washed in a buffer containing antibiotics and antimycotics and separated from the placenta, wherein the amniotic membrane is enriched with mesenchymal stem cells derived from the caprine amniotic membrane by explant culture and/or lyophilized conditioned media derived from said mesenchymal stem cells cultured in serum-free medium. The scaffold may be cryopreserved by vitrification and retains native extracellular matrix components. The disclosure further provides a process for preparation of the scaffold comprising collection of placenta, separation of the amniotic membrane, explant culture to obtain mesenchymal stem cells, collection and lyophilization of conditioned media, and enrichment of the scaffold. The disclosure also provides a bioactive hydrogel formulation comprising lyophilized conditioned media, aloe vera gel, and silk fibroin combined to form a composite hydrogel, and a process for preparation thereof.
