Abstract: The present invention relates to substrate molecules suitable for monitoring enzymatic activity via chemiluminescence. The substrates are provided via a new synthetic method that uses new intermediate products, which are more robust, generate fewer side products, and lead to higher yields. The precursors can be converted to caged luciferins such as peptide-linked aminoluciferins or glycosyl luciferins, useful for assessing enzymatic activity such as protease or glycosidase activity.
