Abstract: ABSTRACT BLOOD AND ANIMAL COMPONENT-FREE MEDIA AND A FERMENTATION METHOD FOR PRODUCING BACTERIAL POLYSACCHARIDES THEREFROM The present disclosure relates to a cost efficient, robust culture media, blood and animal component-free, comprising a carbon source, a nitrogen source, a yeast extract, a phosphate source, salts and microelements, for producing bacterial polysaccharides. The media components provide a synergistic effect which when aligned with pH and OD dependent feeding strategy enhances cell growth in minimum time period, reduces downstream processing challenges, ensures reliable and large-scale production of polysaccharide, providing high yields and low impurities to achieve controlled polysaccharide molecular size and optimal CPS side chain preservation. The invention further discloses a method of growing, fermenting, inactivating bacterial polysaccharide to obtain high yield of purified bacterial polysaccharide. The invention further discloses use of purified bacterial polysaccharide in monovalent or multivalent Streptococcus pneumoniae polysaccharide-protein conjugate vaccine compositions for prevention and/or treatment of subjects with Streptococcus pneumoniae.
