Abstract: ABSTRACT BIOTIN-FUNCTIONALIZED LIPID–POLYMER HYBRID NANOPARTICLE COMPOSITION FOR TARGETED DELIVERY OF DOXORUBICIN TO CANCER CELLS The present invention relates to a biotin-functionalized lipid–polymer hybrid nanoparticle composition (100) for targeted delivery of doxorubicin, comprising lipid–polymer hybrid nanoparticles (102) encapsulating doxorubicin (104), each nanoparticle (102) having a polymeric core (102A) of polyethylene glycol–poly(lactic-co- glycolic acid), a surrounding lipid layer (102B) of phospholipids, and a biotin ligand (102C) conjugated to the polyethylene glycol segment to enable receptor-mediated uptake by cancer cells. The invention further relates to a process (200) for preparing the nanoparticles (102), comprising activating (202) biotin, conjugating (204) the activated biotin with an amine-terminated polyethylene glycol–poly(lactic-co-glycolic acid) polymer to obtain a biotin-PEG-PLGA copolymer, preparing (206) an organic phase containing doxorubicin, the copolymer, and a phospholipid, emulsifying (208) the organic phase with an aqueous phase containing polyvinyl alcohol, subjecting (210) the emulsion to sonication to form nanoparticles (102), and removing (212) the organic solvent.
