Abstract: The present invention relates to the field of medicinal chemistry, specifically to the development of bioisosteric analogues for selective alpha-amylase inhibition, which is crucial in diabetes management. The invention comprises a core structure featuring a bioisosteric analogue of a thiazolidine ring, selected from thiadiazole and oxazole rings, with strategically positioned hydroxyl and carboxyl groups to enhance hydrogen bonding with the active site of alpha-amylase. Additionally, methoxy or ethoxy side chains are attached to the core structure to increase lipophilicity, thereby enhancing binding affinity and reducing degradation in the gastrointestinal tract. The invention also includes methods for preparing these bioisosteric analogues, involving the design of the core structure, incorporation of functional groups through novel catalytic reactions, and evaluation using in vitro enzyme assays and computational docking studies.
