Abstract: ABSTRACT The present invention relates to bioinspired mucus-penetrating nanoparticles for enhanced delivery of therapeutic agents across mucus-covered biological surfaces. The nanoparticles comprise a biocompatible or biodegradable nanoparticulate core containing at least one therapeutic, prophylactic, biological, diagnostic, or imaging agent and a bioinspired hydrophilic surface layer configured to reduce interactions with mucin. The surface may comprise hydrophilic polymers, zwitterionic materials, phospholipids, polysaccharides, glycopolymers, peptides, biomimetic membrane components, or combinations thereof. The nanoparticles preferably possess nanoscale dimensions, controlled particle-size distribution, high surface hydration, and substantially neutral or weak surface charge. The bioinspired surface minimizes hydrophobic, electrostatic, and nonspecific interactions with mucus, thereby facilitating improved diffusion toward the underlying epithelial surface. The nanoparticles may provide protection of encapsulated agents, controlled release, enhanced mucosal transport, and improved epithelial availability. The platform can be adapted for delivery of small molecules, phytoconstituents, peptides, proteins, nucleic acids, vaccines, diagnostic agents, and combinations thereof. The nanoparticles are suitable for nasal, pulmonary, gastrointestinal, buccal, ocular, vaginal, rectal, and other mucosal delivery applications and provide a versatile platform for pharmaceutical and biomedical use.
