Abstract: The present invention relates to a novel biocomposite dental formulation composed of Blepharis maderaspatensis extract, chicken eggshell powder, and carboxymethyl cellulose (CMC), developed for the remineralization of enamel and dentin with adjunct anticariogenic, antimicrobial and bioactive properties. The formulation further demonstrated concentration-dependent biological activity and acceptable cytocompatibility in Saos-2 cells, supporting its safety for the intended dental application. Antimicrobial assays confirmed significant inhibition of Streptococcus mutans and mild antifungal activity from eggshell powder, while Blepharis extract did not exhibit measurable antifungal activity under the experimental conditions. The formulation demonstrated effective remineralization potential via Micro-Vickers hardness testing, SEM–EDS analysis, FTIR Charcterization and Alizarin Red S staining. The optimized biocomposite gel contains Blepharis extract at 0.30% w/v, chicken eggshell powder at 31.2 µg/mL, and CMC at 2.0% w/v, and is prepared without any humectant and without any pH adjustment, being maintained at its inherent pH.
