Abstract: ABSTRACT BIOACTIVE PEPTIDE-LOADED INJECTABLE SELF-HEALING HYDROGEL FOR ENHANCED BONE REGENERATION The present invention relates to an injectable self-healing hydrogel comprising a bioactive osteogenic peptide incorporated within a biocompatible dynamically crosslinked polymeric network for enhanced bone regeneration. The developed hydrogel was prepared by combining biocompatible polymeric components capable of forming reversible crosslinks and incorporating a bioactive peptide having osteogenic activity. The optimized hydrogel demonstrated shear-thinning and injectable characteristics, rapid recovery following mechanical disruption, high swelling capacity and controlled peptide release. Physicochemical and rheological characterization demonstrated formation of a stable three-dimensional hydrogel network. In-vitro studies demonstrated acceptable cytocompatibility, enhanced cellular proliferation, increased alkaline phosphatase activity, enhanced calcium deposition and increased expression of osteogenic markers including RUNX2, ALP, COL1A1, osteocalcin and osteopontin. In an experimental bone-defect model, administration of the peptide-loaded self-healing hydrogel resulted in enhanced new-bone formation, improved bone-volume fraction, mineral density and defect bridging compared with untreated and blank-hydrogel groups. The invention therefore provides an injectable, self-healing and sustained- release biomaterial platform for minimally invasive bone repair and regeneration.
